How much of a difference is there between the yamaha 3,4, and 5 hundred series. Is there really a discernable difference in tone?

The 3 and 4 series, no. The 5 series yes because you get a different cut headjoint on it (the EC) than on the 3 and 4 series which have a CY cut.
